OligoTex Protocol for 1 mg Total Yeast RNA

Prepare in advance:
- 1.5-4 mg Total RNA in 750 µl H2O
- pre-heat oligoTex beads to 37 C, vortex, set at Room Temp
- warm buffer OBB to 37 C, mix, place at Room Temp
- place 150 ul aliquot of OEB buffer at 70 C
mRNA purification:
- prepare 1 mg of Total RNA in 500 ul H20 or TE
- add 500 ul OBB
- add 55 ul OligoTex beads
- heat sample to 70 °C for 3 min.
- place at Room Temp, 10 min.
- spin 2 min., max speed
- remove sup
- re-suspend pellet in 400 ul OW2 by pipetting and move sup into a small spin column on a 1.5 ml tube
- spin 1 min, max speed
- remove eluate
- apply 400 ul OW2 and spin 1 min.
- transfer column to new tube
- apply 50 ul HOT OEB, pipet up and down to re-suspend resin
- spin 1 min.
- apply 50 ul HOT OEB, pipet up and down to re-suspend resin
- spin 1 min.
- place eluate on ice
yield is typically 5 - 15 ug
