Is the surrounding of the microscope clean and organized?

Is the following maerial for microscope maintenance and operation available?

  • Cleaning solution for objectives
    • 85% ether, 15% isopropanol for oil immersion
    • Lens cleaner for air and water immersion
  • Lens cleaning tissue
  • Swabs
  • Dust blower
  • Immersion liquid
    • oil for oil immersion
    • di-water (better: flourescence free water) for water immersion
    • plastic pipettest apply water
  • Manual
  • Approved containers for sharps and biohazard disposal

Is vibration-free worksurface operational? (if available)

Is vibration isolation sufficient?

Performance test
Setup Focus a Fluocell #3 slide using highest magnification objective.
Camera available

Take 60 images 1 sec intervals while walking about the room. Does the movie show vibrations?

Camera not available Observe sample through the eye-pieces while an other person walks about the room. Do not refocus! Does the sample stay in focus? Will you be able to work under these conditions?

Is the microscope surrounding dark enough to work with the scope?

Is a knowledgeable person in charge of microscope?

Do you have any other observations?

Does computer start without error messages?

Is virus scan installed, active and updated?

Is there sufficient free space on the system hard-drive? (recommend 25%)

Is the system hard-drive defragmented?

Check objectives with dissection scope. Clean if necessary.

Are correct objectives assigned to software and hardware?

Are nose piece positions not used closed?

Are the correct DIC sliders or placeholders in place? Are they clean?

After checking other components of microscope, take image of Fluocell #3 slide. If performance is questionable, acquire point-spread function (camera and confocal systems only)

Check these assemblies for contamination, damage and misssing parts:

  • microscope stage
  • eyepieces
  • condenser
  • filters and analyzer in condenser assambly
Performance test
Setup

Focus a convalleria slide using a low magnification objective with Koehler illumination.

bulb alignment Remove eye-pieces and look down beam-path (preferable with phase telescope)
Setup and check other transmitted light contrast methods if applicable.
DIC Remove eye-pieces and look down beam-path
Phase Remove eye-pieces and look down beam-path
